Effect of TLR4 on the expression of IL-1β and TNF-α in SFs. SFs were pre-treatment with or without TAK-242, then stimulated with LPS. Cell lysates were collected (at 12 h) for analyzing the protein expression by Western blot (a), and the relative expression of pro-IL-1β (b) and TNF-α (c) was calculated. Total RNAs were extracted (at 6 h) for detecting mRNAs of IL-1β (d) and TNF-α (e) by real-time quantitative PCR. The cell-free culture medium from each sample was collected at 12 h, and the mature IL-1β (f) and TNF-α (g) proteins secreted into the cell culture media were measured by ELISA. Data shows all the values from independent samples of , versus nonstimulated controls or cells without TAK-242 treatment.
