ASCs and IL-10 regulate the profile of cytokines produced by macrophages (M). (a) M (2 × 10⁶ cells/well) were cultured alone, cocultured directly with ASCs, or cocultured with ASCs via Transwell (M : ASCs ratio = 4 : 1) for 48 h. Cells were then washed and incubated for 24 h with fresh culture medium with or without LPS (10 ng/mL). (b) Neutralizing IL-10 antibody (IL-10Ab, 10 ng/mL), IL-10 receptor antibody (IL-10RAb, 10 ng/mL), or IgG control (10 ng/mL) was added to the medium at the beginning of the Transwell cocultures of ASCs + M. Then, cells were treated with or without LPS (10 μg/mL) for another 24 h. Culture supernatants were harvested for cytokine assay via Luminex.
