LPA-induced DR6 upregulation is specifically mediated by PKCδ isoform. (a) A time course of LPA-induced activation of PKC kinases. The blots were incubated in sequence with antibodies against several major PKC isoforms such as PKCα/β, PKCε, PKCζ/λ, and PKCδ. All isozymes stained as a dominant band of the expected molecular weight. . (b) HeLa cells were infected with adenoviral PKCα-DN, PKCδ-DN, PKCε-DN, and PKCζ-DN for up to 24 hrs. Cells were then starved in DMEM serum-free overnight and stimulated with 25 μM LPA (+) or vehicle (−) for 8 hrs, followed by Northern blotting for DR6 mRNA expression. .
