TSG-6 Downregulates IFN-Alpha and TNF-Alpha Expression by Suppressing IRF7 Phosphorylation in Human Plasmacytoid Dendritic Cells

The suppressive effect of TSG-6 was dependent on CD44. GEN 2.2 cells were treated with anti-CD44 blocking antibody at various concentrations as indicated or with control IgG (mouse IgG2b) for 45 min, followed by TSG-6 (1000 ng/mL) overnight, and stimulation with CpG-A (2 μM) (a) or R837 (10 ng/mL) (b) for 6 hours.
