Human primary trophoblast cells were transfected with 50 nM siKRAS, 50 nM siHRAS, or 50 nM siCONT for 48 h then treated with 10 ng/ml TNF-α for an additional 24 h ( patients). (a–d) IL-6, IL-8, MCP-1, and GRO-α mRNA expression was analysed by qRT-PCR, and the fold change was calculated relative to TNF-α-stimulated siCONT-transfected cells. (e–h) The incubation medium was assayed for concentration of IL-6, IL-8, MCP-1, and GRO-α release by ELISA.
