Region of the Kat6b 3UTR sequence containing the putative miR-487b conserved binding site, indicated in bold. The complete 3UTR (1995 bases) was inserted into the pLightswitch 3UTR luciferase reporter plasmid, using the NHE1/XHO1 restriction sites, generating the pKat6bLuc-3UTR plasmid (SwitchGear Genomics, Carlsbad, CA, Product ID S810637). Five bases (TACGA) were deleted from the miR-487b binding site to generate the mutant clone (pKat6bLucΔ3UTR).
