Effects of Temperature and Water-Filtered Infrared-A Alone or in Combination on Healthy and Glyoxal-Stressed Fibroblast Cultures

Metabolic activity. The bar chart shows metabolic activity of the cells as determined by resazurin conversion assay at different temperatures and after treatment with or without glyoxal and/or wIRA. Squared brackets indicate statistically significant differences ( ) between temperatures. Cells were able to maintain adequate metabolic activity at temperatures up to 42°C, but their metabolic activity declined significantly at 45°C. The micrographs show cells that were stained with JC-1 for visualising the mitochondrial membrane potential. Red fluorescence, as seen in cells cultured at 37°C, indicates a highly negative mitochondrial membrane potential, while green fluorescence, as seen in cells cultured at 45°C, indicates that the mitochondrial membranes were depolarised. No differences between the treatment groups (with or without glyoxal and/or wIRA) could be observed. Scale bar: 100 μm.
