Preconditioning and Acute Effects of Flavonoids in Protecting Cardiomyocytes from Oxidative Cell Death

Effects of 3-day pretreatment with sodium ascorbate (Asc) against ischemia-reperfusion-induced cell death in H9c2 cells. Confluent cells were maintained in DMEM with 2% FBS for 6 days including 3-day pretreatment with different concentrations of ascorbate, and then ischemia-reperfusion groups were exposed to ischemia and reperfusion. Ascorbate was also present during the reperfusion time. Cell viability was determined using a microscope after staining the cells with acridine orange. Bars are means of 4-5 wells ± SEM. *Indicates compared to the nontreated ischemic-reperfused group (IR). There were no significant effects of ascorbate on growth and viability of the cells under the control (non-IR) condition.
