Mtl1 N-glycosylation is required for efficient stress-induced PCD execution. (a) The mtl1Δ strain (RSY1660) transformed with plasmids expressing MTL1 (pCJ4), MTL1 N42A (pCJ10), or the vector control was subjected to H2O2 or acetic acid at the indicated concentrations for 240, 200, and 200 m, respectively. Caspase assays were performed on three independent cultures and the mean ± S.E.M. is indicated. * , ** . (b) Strains described in (a) were grown to midlog phase and then either treated or not with 100 mM acetic acid for 200 min. The cells were serially diluted (1 : 10) and then spotted onto growth medium. The images were obtained following 2 days incubation at 30°C.
