Involvement of PKC in TPA-induced EMT processes in MCF-7 cells. (a) MCF-7 cells were treated with 1 nM TPA for the indicated times. The membrane translocation of phosphorylated PKC was then determined by Western blotting. Values are the mean of fold changes from vehicle-treated cells (, ). The cells were pretreated with 5 μM GF109203X (GF) or 10 μg/mL actinomycin D (ActD) for 1 h and were then treated with 1 nM TPA for 6 h (b) or 24 h (c). RT-PCR was carried out. RT-PCR data were normalized using β-actin levels (, versus vehicle, , versus TPA-treated cells).
