Xanthine Oxidase-Derived ROS Display a Biphasic Effect on Endothelial Cells Adhesion and FAK Phosphorylation

Hx-XO-derived ROS induced time-dependent biphasic HUVEC adhesion. HUVECs monolayers were treated with Hx-XO (4.5 mU/mL) for different time periods and then thoroughly washed to remove nonadherent cells. Cells adherent to the matrix were loaded with 2.5 μM Calcein-AM and fluorescence was analysed in a FLUOstar plate reader (BMG, Lab Technologies, France) using a 480 nm excitation wavelength and recording of green fluorescence at 520 nm. Values were expressed as mFU units per well (a) or as percentage of control cells without Hx-XO (b). Results are expressed as mean ± SD, , versus untreated cells.
