Downregulation of Glutathione Biosynthesis Contributes to Oxidative Stress and Liver Dysfunction in Acute Kidney Injury

Proposed mechanism of kidney ischemia-reperfusion induced reduction of hepatic glutathione biosynthesis. Renal ischemia-reperfusion (IR) inhibits glutathione biosynthesis in the liver through (1) a decrease in the expression of cystathionine γ-lyase (CSE) in the transsulfuration pathway which elevates homocysteine and reduces de novo cysteine production and hydrogen sulfide (H2S) generation and (2) a decrease in the expression of glutamate-cysteine ligase (GCL) which is a key enzyme responsible for glutathione biosynthesis.
