The PPAR𝛿 Ligand GW501516 Reduces Growth but Not Apoptosis in Mouse Inner Medullary Collecting Duct Cells

PGE₂ does not affect PPAR𝛿 mRNA expression. RNA was isolated from IMCD-K2 cells using the TRIzol method. The cells had been treated for 2, 8, and 24 hours with serum-free media (control), 10⁻⁵ M indomethacin (Indo), or 10⁻⁶ M PGE₂ in the presence of 10⁻⁵ M indomethacin (PI). The RNA samples were quantified using real-time PCR using primers and probes for PPAR𝛿. Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) was used as an internal control. Expression is presented as the amount of PPAR𝛿 mRNA expression divided by GAPDH expression. Values are means ±  S.E.M.; 𝑛=5.
