Sonic Hedgehog Produced by Bone Marrow-Derived Mesenchymal Stromal Cells Supports Cell Survival in Myelodysplastic Syndrome

Cyclopamine and demethylating agent 5-aza-dC synergize to induce survival inhibition of MUTZ-1 and SKM-1. MUTZ-1 cells were treated with a combination of cyclopamine and 5-aza-dC or with either agent alone in the presence or absence of Shh-N for 48 h. Cell proliferation and apoptosis were determined using CCK8 assay (a) and Annexin V-PI staining analysis (b), respectively. . Flow cytometric data are shown from one representative experiment.
