Human Bone Marrow Mesenchymal Stem/Stromal Cells Preserve Their Immunomodulatory and Chemotactic Properties When Expanded in a Human Plasma Derived Xeno-Free Medium
Figure 3
Motility of hMSC cultured in XF-Medium. (a-b) Recruitment assays—the lower compartment was filled with serum-free medium (negative control) or with medium supplemented with 30% FBS (positive control); hMSC were expanded in XF-Medium or FBS-Medium (, Mann-Whitney test). (c–e) Time-lapse analysis—hMSC expanded in FBS-Medium or XF-Medium were incubated overnight in DMEM without supplements. Snapshots were taken every 10 minutes for 12 hours (c). The percentage of motile cells was calculated counting the number of motile cells and the total number of cells that appear in each video (; unpaired t-test) (d). The velocity of 3 randomly selected cells in each video was calculated using Fiji program. The average velocity from hMSC expanded in FBS-Medium and XF-Medium is shown (; Mann-Whitney test) (e).
