The Exposure to Osteoarthritic Synovial Fluid Enhances the Immunomodulatory Profile of Adipose Mesenchymal Stem Cell Secretome

The conditioned medium of SF-exposed AMSCs inhibits the proliferation of CD3+ T cells. Gating strategies of T cells in proliferation assay—physical parameters, i.e., forward scatter (FSC) and side scatter (SSC), were used to select PBMCs. T cells were recognized by evaluating in PBMCs the expression of CD3. Proliferation was expressed as fold change of the proliferative cells with respect to stimulated control cells (a). CFSE-labelled PBMCs isolated from healthy donors were incubated in the presence of conditioned medium of unstimulated (CM) or SF-exposed AMSCs (CM+SF) or synovial fluid only (SF).
