The conditioned medium of SF-exposed AMSCs increased the expansion of T reg. CD4+ T cells, isolated from PBMCs by negative selection, were stimulated with anti-CD3, anti-CD28, and IL-2 in the presence of conditioned medium of untreated cells (CM), conditioned medium of AMSCs exposed to synovial fluid (CM+SF), and synovial fluid only (SF). The percentage of regulatory T cells (CD4+/CD25+/FoxP3+) was determined by fluorescence-activated flow cytometry (FACS) on day 4.
