Size-Optimized Microspace Culture Facilitates Differentiation of Mouse Induced Pluripotent Stem Cells into Osteoid-Rich Bone Constructs

<div>Microspace size influences the osteogenic marker gene expression of OI-iPSC constructs. The expression of osteogenic marker genes in Elp400, Elp500, and Elp900 was determined by real-time RT-PCR. (a and b) The gene expression of osteogenic-related transcriptional factors including (a) <i>Runx2</i> and (b) <i>Osterix</i> (<i>Osx</i>) was determined at day 10 of osteogenic induction. (c–f) After maintenance for 20 days, the expression of bone ECM-related genes-related genes such as (c) <i>Collagen 1a1</i> (<i>Col1a1</i>), (d) <i>Bone sialoprotein</i> (<i>Bsp</i>), (e) <i>Osteopontin</i> (<i>Opn</i>), and (f) <i>Osteocalcin</i> (<i>Ocn</i>) was evaluated. <i>18</i>s <i>rRNA</i> expression was used as an internal control.
