Melatonin Alleviates LPS-Induced Pyroptotic Cell Death in Human Stem Cell-Derived Cardiomyocytes by Activating Autophagy

Figure 3

Melatonin improves cell viability in LPS-treated hiPSC-CMs. (a) hiPSC-CMs were challenged with various doses of melatonin (0.1, 1, 10, and 100 μmol/l) for 12 h; the viability of hiPSC-CMs was measured by CCK-8 assay. (b) hiPSC-CMs were treated with various doses of LPS (0.1, 1, 2.5, or 5 μg/ml) for 24 h. (c) hiPSC-CMs were incubated with LPS (1 μg/ml) for 24 h post melatonin pretreatment (0.1, 1, 10, or 100 μmol/l) for 12 h; then, the cell viability of hiPSC-CMs was assessed by CCK-8 assay. (d) Measurement of LDH release level in the same condition of (c). ; vs. control; vs. control; # vs. LPS; ## vs. LPS.
