Development of a FLIPR Assay for the Simultaneous Identification of MrgD Agonists and Antagonists from a Single Screen

Figure 5

Illustration of the dose-response curve of selected agonists and antagonists. An agonist identified, ( ± ) HA 966, is a racemic mixture, and further characterization of the positive and negative enantiomers showed that the latter was the most potent MrgD agonist among all three (Figure 4(a)). The EC50 values were 11, 106, and 5.5  𝜇 M for the racemic mixture, positive, and negative isoforms of the compound, respectively. Figure 4(b) shows two representative MrgD antagonists thioridazine and chlorpromazine, with IC50 values of 1.5 and 2.7  𝜇 M, respectively.
