The effect of pyruvate cycling flux on the enrichment of acetyl-CoA from [1-13C]glucose. For clarity, the unlabeled pyruvate that is also generated is not shown. Pyruvate carboxylase converts [3-13C]pyruvate to [3-13C]oxaloacetate which is in rapid exchange with malate and fumarate. This results in a randomization of the 13C label between carbons 2 and 3 of these intermediates and the subsequent formation of [2-13C]pyruvate and [1-13C]acetyl-CoA isotopomers, shown in red.
