Tumor acidosis causes chemoresistance against weak-base drugs. (a) Uptake and retention of mitoxantrone, a weak-base drug, was greater in C3H tumor tissue that was neutralized with sodium bicarbonate. (b) Mitoxantrone treatment (1 dose of 12 mg/kg or 2 doses of 6 mg/kg) caused a greater growth delay when sodium bicarbonate neutralized the tumor acidosis.
