Synthesis of derivatives with a branched alkyl side chain consisting of 5 carbon atoms (5, 7 to 10) and the 4-amino-7-chloroquinolinyl substituted derivatives (11 and 12) with antileishmanial activity. Reagents and conditions: (a) Boc2O, MeCN (dry), 0–25°C; (b) (rac)-1,4-dibromopentane, NaH, DMF (dry), 0–25°C; (c) NaN3, DMF (dry), 25°C; (d) TFA, DCM, 25°C; (e) PPh3, MeOH (dry), 25°C; (f) 4,7-dichloroquinoline, Pd2(dba)3, ±-BINAP, KOtBu, 1,4-dioxane (dry), 85°C; (g) TFA, DCM, 25°C.
