Titration of the recombinant adenovirus: The plasmid, CMV5(Cuo)-IRES-GFP (Qbiogene) containing the iNOS was used to construct and plaque purify the recombinant Ad-hiNOS-GFP adenovirus. The viral plaques were screened by Western blot analysis for expected iNOS detection followed by amplification on 3.0×10⁹ 293 CymR cells and purification on CsCl gradients. The recombinant adenovirus was tittered by the optical density and by TCID₅₀ methods. A sterile test, detection of mycoplasma, endotoxin, and RCAs were also performed.
