Titration of the recombinant adenovirus: The plasmid, CMV5(Cuo)-IRES-GFP (Qbiogene) containing the iNOS was used to construct and plaque purify the recombinant Ad-hiNOS-GFP adenovirus. The viral plaques were screened by Western blot analysis for expected iNOS detection followed by amplification on 293 CymR cells and purification on CsCl gradients. The recombinant adenovirus was tittered by the optical density and by TCID50 methods. A sterile test, detection of mycoplasma, endotoxin, and RCAs were also performed.
