Effects of hBD-3 on the mitochondrial membrane potential change. Neutrophils (106 cells/mL) were incubated at 37°C for 18 h in RPMIFBS in the absence ((b); Control) or presence of μg/mL hBD-3 ((c), 5 μg/mL). Neutrophils were also incubated at 4°C for 18 h in the absence of hBDs ((a); Resting). After incubation, cells were harvested and stained with a mitochondrial membrane potential-dependent lipophilic dye, JC-1. The stained cells were analyzed for the intensities of green fluorescence (FL1) and red fluorescence (FL2) by flow cytometry. Results were expressed as a percentage of cells with disrupted (enhanced green but diminished red fluorescence) or intact (enhanced red but diminished green fluorescence) mitochondrial membrane potential (d). Data are the mean ± SD of 9 to 12 separate experiments. Values are compared between the incubation at 37°C in the absence (Control) and presence of μg/mL hBD-3. *; ***.
