Pharmacokinetics and Biodistribution Study of 7A7 Anti-Mouse Epidermal Growth Factor Receptor Monoclonal Antibody and Its Fragment in an Immunocompetent Mouse Model

Plasma clearance of 7A7 mAb and 7A7 F(ab′)2 fragment in C57BL6 mice. 7A7 mAb (2.8 mg per Kg) or 7A7 F(ab′)2 fragment (1.86 mg per Kg) were injected i.v. into C57BL6 mice. Blood samples were collected at time intervals (1, 10, and 30 min and 1, 4, 8, 12, 24, and 48 h post-injection). Concentration of both molecules in plasma samples was measured using an ELISA system using several plasma dilutions. Absorbance units were converted to μg/mL of mAb or F(ab′)2 fragment using standard curves. Plotted values represent mean of 3 animal’s data points per group ± standard deviation (SD).
