Sustained Release of Protein Therapeutics from Subcutaneous Thermosensitive Biocompatible and Biodegradable Pentablock Copolymers (PTSgels)

(a) In vitro dissolution/disintegration of 10GH PTSgel. Three concentrations (12.5, 18.75, and 25%) of PTSgel 10GH (500 μL in triplicate) were evaluated for disintegration in phosphate buffered saline (pH 7.4) in vitro at 37°C. On day 0 and after 5, 15, 30, and 45 days and every 15 days thereafter, overlaid buffer was removed and the remaining PTSgels were lyophilized and weighed to determine PTSgel dry weight over time. (b) GPC analysis of residual polymer (10GH, 25%) and supernatant collected on various days for in vitro disintegration in PBS buffer, pH 7.4 at 37°C.
