Mycophenolic Acid Overcomes Imatinib and Nilotinib Resistance of Chronic Myeloid Leukemia Cells by Apoptosis or a Senescent-Like Cell Cycle Arrest

MPA induced apoptosis in CD34 CML cells. Primary CD34 cells isolated from blood samples of CML patients responding to imatinib (patient 1) or resistant to imatinib and nilotinib (patient 2 to 4) were grown in the presence of vehicle only, imatinib 1 μM, or MPA 3 μg/mL for 3 days. Patient 2 and 3 overexpressed several tyrosine kinase such as Src-kinase, whereas patient 4 was mutated in Abl kinase domain harbouring the T315I mutation. Annexin-V binding was detected at day 0 and upon treatment at day 3.
