Transcription Factor EB Is Selectively Reduced in the Nuclear Fractions of Alzheimer’s and Amyotrophic Lateral Sclerosis Brains

TFEB protein levels are not reduced in the cytosolic fractions of AD brains. Homogenates were prepared from the hippocampus of AD brains classified as Braak stages II to VI or age-matched normal controls (NC), and cytosolic fractions were separated and subjected to SDS-PAGE. Cytoplasmic fractions showed significantly increased normalized (to tubulin) levels of TFEB protein only at Braak stage IV (61%). Tubulin was detected to ensure equal loading of samples and as a marker for cytosolic fractions. by ANOVA followed by Dunnett multiple comparisons test.
