Jaceosidin Induces Apoptosis in Human Ovary Cancer Cells through Mitochondrial Pathway

Jaceosidin dose-dependently (a) induces loss of mitochondrial membrane potential and (b) induces cytochrome c to release from mitochondria to cytosol. CAOV-3 cells were seeded into 12-well-plate and cultured overnight. Jaceosidin or quercetin was added into each well. Twenty-four hours later, cells were harvested for measuring mitochondrial membrane potential by flow cytometry using JC-1 staining. Cells on the lower side showed lower membrane potential. One of three independent experiments is shown. In addition, mitochondria and cytosol proteins were prepared from cells treated with 80  M jaceosidin, which were subjected to western blotting for measuring cytochrome c in mitochondria and cytosol.
