Efficient siRNA Delivery by the Cationic Liposome DOTAP in Human Hematopoietic Stem Cells Differentiating into Dendritic Cells
Effects of siRNA delivery agents on cytotoxicity and transfection rates. Freshly isolated HSCs were subjected to agent-specific transfection protocols using a fluorescein-labeled, nontargeting siRNA control as described in the Materials and Methods. After 24 hours incubation, cytotoxicity was assessed through the trypan blue dye exclusion test (a), while transfection rates were calculated from the ratio between fluorescein-labeled and total number of DAPI-stained cells (b).
