A Multifactorial Mechanism in the Superior Antimalarial Activity of -C-GalCer

Mechanism of 𝛼 -C-GalCer’s enhanced therapeutic effect against malarial liver stages: ( 1 ) 𝛼 -C-GalCer presented by CD1d molecules expressed on APCs stimulates, ( 2 ) enhanced (+) proliferation by V 𝛼 14i NKT cells relative to 𝛼 -GalCer, and ( 3 ) enhanced (+) activation and IL-12 release by APCs compared to 𝛼 -GalCer, which ( 4 ) induces augmented IFN- 𝛾 production by NK cells. This increased IFN- 𝛾 is responsible for ( 5 ) the better protection against malarial liver stages exhibited by 𝛼 -C-GalCer.
