Cell Density Plays a Critical Role in Ex Vivo Expansion of T Cells for Adoptive Immunotherapy

ATCs proliferate at high cell density. ATCs were activated at high density and then reseeded in fresh growth medium plus IL-2 at varying concentrations. (a) Numbers of ATCs at different cell densities. ATCs were cultured at high cell density ( 1 × 1 0 5 /mL) or low cell density ( 1 × 1 0 4 /mL) in 75 cm2 flasks for 6 days. Cell number change rate (%) was monitored by direct cell counting. (b) Cell division of ATCs at different cell densities. After labeling with CFSE, ATCs were cultured at 1 × 1 0 5 /mL, 1 × 1 0 4 /mL, and 1 × 1 0 3 /mL in 75 cm2 flasks for 6 days. Cell division was monitored by CFSE dilution in the dividing cells as analyzed by flow cytometry.
