Intranasal Immunization with Chitosan/pCAGGS-flaA Nanoparticles Inhibits Campylobacter jejuni in a White Leghorn Model

Protection of DNA from DNAse I digestion by chitosan. Naked DNA and chitosan-DNA complex containing 1โ€‰ ๐œ‡ g DNA were both incubated with DNAse I for 15โ€‰min at 3 7 โˆ˜ C . Then the reaction was stopped by adding with 0.5โ€‰M EDTA. chitosan-DNA was then subjected to chitosanase and lysozyme digestion. Finally, all samples were run on 0.8% (w/v) agarose gel and stained with ethidium bromide. Lane 1: chitosan-DNA; 2: chitosan-DNA + DNAse I (28โ€‰U/mL); 3: chitosan-DNA + chitosanase (0.15โ€‰U/mL) + lysozyme (15โ€‰U/mL); 4: 3 + DNAse I (28โ€‰U/mL); 5: DNA+ DNAse I (28โ€‰U/mL); 6: DNA (pCAGGS-flaA, 4.1โ€‰kb).
