Southern blot analysis for the detection of HBV sequences in human cell lines. The cell lines HEP-3B, HEP-G2, and SK-HEP-1 are cell lines established from human hepatocellular carcinomas. The ARH-77 is a human hematopoietic cell line and serves as a negative control. Genomic DNA was digested with BamHI, separated on an agarose gel, blotted and hybridized with a labeled PCR fragment covering the Hepatitis B surface antigen coding region. The hepatocellular carcinoma cell lines SK-HEP-1 and HEP-G2 display no HBV sequences, whereas HEP-3B shows a hybridization signal at about 1.6 kb originating from HBV sequences integrated into the cellular genome.
