Characterization of Dendritic Cell and Regulatory T Cell Functions against Mycobacterium tuberculosis Infection

Intracellular survival of H37Rv inside mature DCs. Monocyte derived DCs were infected with processed H37Rv at a multiplicity of infection of 1 : 1 (1 bacterium per DC). Infected DCs were cultured in medium containing LPS (1 μg/mL) in the presence and absence of NAC (5, 10, and 20 mM), L-GSH (5, 10, and 20 μM), or BSO (500 μM). Infected DCs were terminated at 1 hour and 5 days after infection to determine the intracellular survival of H37Rv in mature DCs. Results were analyzed using one-way ANOVA with Dunnett’s multiple comparisons test, comparing all treatment categories to the infected-LPS treated control, ^∗𝑃≤0.05, ^∗∗𝑃≤0.01.
