Effect of CBHA on TNF-α-induced activator protein (AP)-1 activity and PAI-1 promoter in MeT-5A cells. (a) and (b) Cells were pretreated with vehicle or CBHA (0.5–1 μM) for 15 min, followed by treatment with TNF-α (10 ng/mL) for 60 min. The nuclear amount of phosphorylated (a) ATF-2 and (b) c-Jun was detected by Western blot analysis of nuclear extracts (NE) with specific antibodies. (c) Cells were transfected with the specific PAI-1 reporter plasmid (p800Luc), together with internal plasmid (Renilla). Transfected cells were pretreated with vehicle or CBHA (0.5–2 μM) for 15 min and then stimulated with TNF-α (10 ng/mL) for 24 h. The luciferase activity of PAI-1 reporter gene was assessed.
