The Proteome of the Differentiating Mesencephalic Progenitor Cell Line CSM14.1 In Vitro

Results from ICC-staining of CSM14.1 cells during differentiation are shown. Images do not represent counting frame pictures and the numbers and distribution of immunoreactive cells should not be compared with the stereological results. However, morphological changes and different contents of cell type specific markers are recognizable. In the first row ((a), (b), (c)) images of staining against nestin (red) and DAPI (blue) are merged. A significant decrease of nestin-expression between undifferentiated cells (a) and cells that differentiated for 14 days (b) or 28 days (c) could be observed. In the second row ((d), (e), (f)) images of staining against GFAP (red) and DAPI (blue) are merged. Undifferentiated cells do not express GFAP (d). After 14 days of differentiation GFAP-positive cells could be detected (d) and an increase of positive cells could be shown after 28 days of differentiation (f). In the third row ((g), (h), (i)) images of staining against the neuronal marker NeuN (red) and DAPI (blue) are merged. No NeuN-positive cells could be detected in undifferentiated cells (g), whereas NeuN expressing cells could be found after 14 days of differentiation (h) and an increase in NeuN-positive cells could be observed after 28 days of differentiation (i). In the fourth row ((j), (k), (l)) images of staining against TH (red) and DAPI (blue) are merged. Undifferentiated cells do not express TH (j). TH-positive cells could be found after 14 days of differentiation (k) and an increase of TH-expressing cells was observed after 28 days of differentiation (l). Scale bars = 200 μm ((a)–(l)).
