Silibinin Inhibits ICAM-1 Expression via Regulation of N-Linked and O-Linked Glycosylation in ARPE-19 Cells

Inhibitory effects of silibinin on NF-κB and STAT1 signaling pathways. ARPE-19 cells were pretreated with 25 μM or 50 μM silibinin under serum-free conditions for 24 h and incubated for a further 30 min in the presence or absence of TNF-α (a) or IFN-γ (b). Lysates of cells stimulated with TNF-α were blotted with an antibody against the p65 subunit of NF-κB (nucleus) and an anti-phospho-IκB antibody (cytoplasm). Lysates of cells stimulated with IFN-γ were blotted with anti-phospho- or anti-total STAT1 antibodies.
