KML001, a Telomere-Targeting Drug, Sensitizes Glioblastoma Cells to Temozolomide Chemotherapy and Radiotherapy through DNA Damage and Apoptosis

DNA damage and apoptosis were enhanced by in vitro combination treatment of KML001 and TMZ. (a) Phosphorylated -H2AX was detected by immunocytochemistry after 40 minutes treatment with 200 M TMZ and KML001 [10 M (for U251MG) or 5 M (for U87MG)]. DAPI (blue) = nuclei. (b, c) U251MG (b) and U87MG (c) cells were treated in vitro as indicated for 48 hours. Western blot analysis for PARP, cleaved PARP, or cleaved caspase-3 was performed. -actin = loading control.
