Rosiglitazone Regulates Anti-Inflammation and Growth Inhibition via PTEN

Rosiglitazone did not block LPS-induced iNOS upregulation/NO release and COX-2 upregulation/PGE2 production in shPTEN cells. Cells were pretreated with 25 M rosiglitazone prior to stimulation with 5  g/mL LPS for indicated times. (a) PTEN protein expression in RAW 264.7 cells transduced with shLuc and shPTEN. Effects of rosiglitazone on the expression of iNOS (b) and COX-2 (c) in shLuc and shPTEN cells. Cell lysates were harvested for detection of levels of PTEN, iNOS, COX-2, and -actin by Western blotting. Cell supernatants were collected for determining NO release and PGE2 production. Data shown are representative of three individual experiments. Data are expressed as mean SEM obtained from three individual cultures. < 0.05, < 0.01, and < 0.001 compared with the control, untreated group. < 0.05 and < 0.01 compared with the LPS group.
