Rosiglitazone did not block LPS-induced iNOS upregulation/NO release and COX-2 upregulation/PGE2 production in shPTEN cells. Cells were pretreated with 25 M rosiglitazone prior to stimulation with 5 g/mL LPS for indicated times. (a) PTEN protein expression in RAW 264.7 cells transduced with shLuc and shPTEN. Effects of rosiglitazone on the expression of iNOS (b) and COX-2 (c) in shLuc and shPTEN cells. Cell lysates were harvested for detection of levels of PTEN, iNOS, COX-2, and -actin by Western blotting. Cell supernatants were collected for determining NO release and PGE2 production. Data shown are representative of three individual experiments.
