The effects of TsES on peritoneal macrophages in septic patients. Macrophages were collected from peritoneal lavage or abdominal cavity flushing fluid in 27 septic patients and 16 control patients and then treated with TsES (5 μg/mL) or the same volume of PBS. Membrane protein fractions were extracted after 6 h in culture. The expressions of TLR2 and TLR4 were detected by western blot (a). The cytoplasmic proteins were extracted after 6 h in culture. MyD88 expression in macrophages was detected by western blot (b). NF-κB activity in nuclear extractions was determined using the NF-κB p50/p65 Transcription Factor Assay Kit (c). Culture supernatants were collected after 6 h in culture. Supernatant TNF-α, IL-1β, IL-10, and TGFβ1 levels were detected by ELISA (d).
