(a) HEK293 cells were transfected with cDNA coding for human MBP with C-terminal FLAG epitope (hMBP-FLAG). After 4 h, the cells were incubated for an additional 20 h with or without GA, as indicated. The cells were then subjected to a cycloheximide chase followed by Western blotting. (b) The HEK293 cells were transfected with cDNAs coding for rhMBP-FLAG along with cDNA coding for the human histone H1.3-FLAG, as indicated. After 24 h, the cells were subjected to a cycloheximide chase followed by western blotting analysis. The percentage of protein remaining was calculated as the ratio of protein at the indicated time-points relative to the initial protein.
