Lunasin Inhibits Cell Proliferation via Apoptosis and Reduces the Production of Proinflammatory Cytokines in Cultured Rheumatoid Arthritis Synovial Fibroblasts

Effect of lunasin on the production of proinflammatory cytokines and MMPs. Synovial fibroblasts in the presence of IL-1β induction were treated with lunasin at various concentrations (0, 10, 50, 100, and 200 μM) for 72 h in serum-free DMEM medium. After treatments, the cell-free medium and RA synovial fibroblasts were collected to evaluate the secretion of IL-6 (a), IL-8 (b), MMP-1 (c), and MMP-3 (d) by their corresponding ELISA kits, respectively. (e) The gene expression of IL-6, IL-8, MMP-1, and MMP-3 was examined by RT-PCR. Data were expressed as mean ± SD for three independent experiments by GraphPad Prism Software Version 5.0. was considered a significant difference when compared with the control.
