PTEN Inhibits Cell Proliferation, Promotes Cell Apoptosis, and Induces Cell Cycle Arrest via Downregulating the PI3K/AKT/hTERT Pathway in Lung Adenocarcinoma A549 Cells

PTEN negatively regulates hTERT by inhibiting the PI3K/AKT pathway. Untransfected A549 cells (con) or A549 cells transfected with either pGFP (null), pGFP-PTEN (wild-type PTEN), pGFP-PTEN-C124A (phosphatase-dead PTEN), or PTEN-siRNA and/or exposed to the AKT inhibitor (LY294002 10 M) were examined for the expression of PTEN (a) and hTERT (b) mRNA by RT-PCR, as well as for the protein levels of PTEN, hTERT, and p-AKT (c) by Western blot.
