Effect of anti-IgM antibody on cell proliferation using competition assay. (a) Immunofluorescence analysis (magnification = 200x): various doses of anti-IgM (0, 10, and 100 μg/mL) were added to samples containing 100 μg/mL of FITC-labeled or unlabeled LG. We observed that anti-IgM antibody could compete with FITC-labeled LG for binding on the cell surface receptor; (b) MTT assay: samples from the receptor binding competition assay, LG progressively lost its ability to stimulate cell proliferation in a dose-dependent manner ().
