Preosteoclast. The interaction between RANK and RANKL induces the recruitment of TRAF6 leading to the activation of extracellular regulated kinase (ERK), p38 mitogen-activated protein kinase (p38), and c-jun N-terminal kinase (JNK). Therefore, transcription factors such as AP-1 and NF-B are activated and subsequently NF-B and NFATc2 are recruited to the promoter of NFATc1, which has been called the master regulator of osteoclastogenesis.
