Proliferative and Invasive Effects of Progesterone-Induced Blocking Factor in Human Glioblastoma Cells

PIBF increases proliferation and the number of U87 cells. Cells were treated with recombinant PIBF (100 and 200 ng/mL) during 5 days, while the medium was used as the vehicle (V). Cells were harvested each day to determine the number of cells (a) and the cell viability (b) by using the trypan blue dye exclusion method. Cell proliferation was measured after the treatment of PIBF (100 ng/mL) during 4 days. Representative immunofluorescence images of cell nuclei (Hoechst stain, upper panel), BrdU positive cells (middle panel), and the merged image (lower panel) are shown (c). The graph shows the percentage of cells incorporating BrdU (d). The data were obtained from three independent experiments and are expressed as the mean ± S.E.M. versus vehicle.
