Natural Compound Oridonin Inhibits Endotoxin-Induced Inflammatory Response of Activated Hepatic Stellate Cells

Oridonin inhibits LPS-induced pro-inflammatory cytokines and cell adhesion molecules. LX-2 cells were pretreated with oridonin (2.5, 5, or 7.5 μM) for 1 h, then stimulated with LPS (1 μg/mL) for 24 h. Cell culture medium was collected for ELISA of IL-6 (a) and MCP-1 (b). Cells were pretreated with oridonin at different concentrations for 1 h and then incubated with LPS (1 μg/mL) for 7 or 9 h and total proteins were examined by Western blots with antibody of IL-1β (c), or ICAM-1, VCAM-1 (d). GAPDH was used as internal control. Densitometric analyses of bands were quantified and data expressed as fold of control normalized to GAPDH.
