Natural Compound Oridonin Inhibits Endotoxin-Induced Inflammatory Response of Activated Hepatic Stellate Cells

Oridonin suppresses LPS-induced NF-κB p65 nuclear translocation and DNA binding. LX-2 cells were pretreated with 7.5 μM of oridonin for 1 h, then stimulated with LPS (1 μg/mL) for 30 min. Nuclear protein was isolated for Western blot assay using NF-κB p65 antibody. Lamin A and Topo βII were used as internal control to normalize the nuclear p65 protein (a). The subcellular translocation of NF-κB p65 was imaged by immunofluorescence assay (b). DNA binding activity of NF-κB p65 was analyzed by TransAM p65 ELISA kit with 10 μg of nuclear proteins (c) and 20 pmol of the wild-type NF-κB oligonucleotide as competitor (lane 4).
